The copy-number of a gene for a given strain used in the co-cultivation experiments was evaluated and compared to the growth rate of each individually cultivated microorganism. qPCR was performed using the CFX96 system (BioRad, CA, USA). Primers used in this study are listed in Table S2. Each PCR reaction mix contained the following: 7.5 μl 2× SYBR SuperMix Green (BioRad, CA, USA), 5 μl of DNA dilution, each of the forward and reverse primers at 0.5 μM and nuclease-free water was added to obtain a final volume of 15 μl. PCR products were detected with SYBR Green fluorescent dye and amplified according to the following protocol: one cycle of 95 °C for 3 minutes, followed by 39 cycles of 95 °C for 5 s and 60 °C for 20 s. Melting curve: 65 °C to 95 °C with increments of 0.5 °C/s. In each run, negative controls (no DNA) for each primer set were included. Standard curve was built using the CFX96 software (BioRad).
